What format does IDT require for oligo pool orders?
IDT oPools require a CSV or Excel file with columns: Pool Name, Sequence Name, Sequence (5' to 3'). Additional optional columns include Scale (typically "oPool" for pools), and Notes. Sequences must be 15-350 nt for oPools (standard) or 15-200 nt for oPools with modifications. IDT individual plates require: Well Position, Name, Sequence, Scale (25nm/100nm/250nm/1um/10um), Purification (STD/HPLC/PAGE). Our adapter handles both formats.
What are the sequence length limits for each vendor?
IDT oPools: 15-350 nt. Twist Oligo Pools: 15-300 nt (standard), up to 350 nt with extended Oligo Pools. GenScript standard oligos: 5-100 nt. GenScript gene fragments: 100-3000 bp. Can I include modifications in my vendor order?
Yes, but modification support varies by vendor and product line. For IDT: standard modifications (5'-phosphate, biotin, fluorophores) are supported for individual oligos but not for oPools. For Twist: oligo pools are unmodified only. For GenScript: extensive modification catalog for individual oligos. Our adapter includes modification fields where supported and warns when modifications are not available for the selected product type.
What happens if my sequence contains invalid characters?
Our adapter validates all sequences against the target vendor's accepted alphabet. Most vendors accept standard DNA bases (A, T, C, G) and some accept IUPAC ambiguity codes (R, Y, S, W, etc.) depending on the product type. Sequences containing invalid characters (numbers, spaces, non-IUPAC letters) are flagged with specific error messages. You can fix these in the tool before downloading the order file, preventing submission errors and order delays.
